It depends on what line you are looking for. G1 or newer or JPN?

Since we're asking... anyone know where I might find the deluxe classics line? I'm looking for Grimlock, but wouldn't mind Starscream or Skywarp.

If you are looking for the originals then eBay would be your best bet. JPN reissue line is available from Silver Snail but you will need to ask them about ordering them and it could get expensive. I would try eBay on that front as well.

You can try Toys R Us stores for Grimlock and Starscream or there is always eBay again. Might be hard to find now since they have been out for a while and had a few sales on them over the last 6-8 months. You might get lucky. Skywarp was only available in a US TARGET exclusive set alongside Ultra Magnus as shown below:
