From what I understand this game is going to be twice the size of Brotherhood. Desmond is also getting a lot more game time and missions.
Ezio's new look is awesome too. I love how they blend his traditional renaissance look with that of the middle east.

Yeah, apparently Ezio gets an ancient device that's pretty much the Animus, and uses that to play as Altair the same way Desmond would play as the both of them. Also Desmond is still playable.
I still have to play Brotherhood...I also have to play Assassin's Creed 2, actually.
