It's a hilarious line, but I have no idea where it comes from. Tuturro says it, and no one reacts to it. It's like everyone in the SUV heard it and just ignored him because he's nutz.

Turturro is well-known as somebody who makes up his own lines. I saw a (rare) interview with the Coens and they said that pretty much 90% of his lines in The Big Lebowski were just ad-libbed on the spot.

Megs got some of the best lines of the movie. Here's another one:

Megatron: You still fight for the weak, that is why you lose!

I actually quote Transformers quite a lot, which is pretty sad.
Most have already been mentioned, but I don't think these two have:

That movie has some of the best quotes ever. Especially if Optimus or Megatron says it.
