According to the Bible and the script for the 86 TF movie Daniel was about 5 years younger then Spike was when we first meet him in the G1 toon.
Now trying to figure out Spike's age seems difficult but in more then one episode of G1 he makes there are refrances to the fact that he's to young to drive.One time by Spike himself in the first season and the other by a police officer in the second season.
Now it is my understanding that the legale age for driving on the West Coast back in 1984 was 16 or 17 years old.
So for Spike not to be old enough to drive he had to be at least 14 years old when we first meet him [and probably younger based on the cop's reaction].
So if you subtrack about 6 years from the age Spike must be in the first season of G1 you end up at 7 years of age.
So G1 Daniel was about 8 years old in the 86 movie.Far closer to what you thing Animated Daniel looked like.
This makes a bit more sence then Daniel being 12 for other reasons.At the time of the movie [1986] the ideal family starting age for a young man was considered to be between 26 and 27 years of age.But if Daniel was 12 like you suggest [and I'll admit he looked] he would have had to been born when Spike was 22 or 23 years of age.
Thats a bit to young by the moral standereds or the time.
Now I know people were having kids at younger ages then, I my self had kids at the age of 13 but I doubt that a 80's cartoon would have been advocating anything less then what the U. S. govererment would have considered the ideal family starting age.

Now Professor Sumdac seemed to be liveing alone so I'll guess his age to be somewhere between 18 and 21. That would place his older self at about 68 to 71.
I saw nothing that suggests that he was living alone and he looked to me to be about 15 or 16 years old.

Not by todays standereds.more then half of my youngest daughter's friends parents are all in there late 60's to mid 70's.
Granted most have older children too.
And besides my mother had me in her late 50's and my father was already 61 at the time and that was back in 1971.
